A study by Katie Lee, a <abbr title=\"doctor of philosophy\">PhD<\/abbr> candidate in the <a href=\"http:\/\/jabsom.hawaii.edu\">John A. Burns School of Medicine<\/a> (<abbr>JABSOM<\/abbr>), explores the role of the Pyruvate kinase <abbr>M2<\/abbr> (<abbr>PKM2<\/abbr>) enzyme in the heart\u2019s energy production and stress management. While previous studies have focused on PKM2 in disease contexts, Lee&#8217;s work explores its function in healthy hearts.<\/p>\n<p>&ldquo;People have explored <abbr>PKM2<\/abbr> in the context of disease,&rdquo; said Lee. &ldquo;The disease has already happened, and they&#8217;re looking at what can be done with <abbr>PKM2<\/abbr> to solve it. We wanted to see what <abbr>PKM2<\/abbr> actually does in healthy hearts, and we discovered that your heart can&#8217;t really be healthy without <abbr>PKM2<\/abbr>.&rdquo;<\/p>\n<h2>Energy production, stress management<\/h2>\n<p>The study examined mice lacking <abbr>PKM2<\/abbr> and found a significant depletion of Adenosine triphosphate, the primary energy source at the cellular level. Lee\u2019s research highlights that, without <abbr>PKM2<\/abbr>, glucose intended for energy production is diverted to manage oxidative stress\u2014a harmful condition driven by reactive oxygen species.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/www.hawaii.edu\/news\/wp-content\/uploads\/2024\/09\/manoa-jabsom-katie-lee-2-300x169.jpg\" alt=\"lee working in the lab\" width=\"300\" height=\"169\" class=\"alignright size-medium wp-image-203722\" srcset=\"https:\/\/www.hawaii.edu\/news\/wp-content\/uploads\/2024\/09\/manoa-jabsom-katie-lee-2-300x169.jpg 300w, https:\/\/www.hawaii.edu\/news\/wp-content\/uploads\/2024\/09\/manoa-jabsom-katie-lee-2-130x73.jpg 130w, https:\/\/www.hawaii.edu\/news\/wp-content\/uploads\/2024\/09\/manoa-jabsom-katie-lee-2.jpg 676w\" sizes=\"auto, (max-width: 300px) 100vw, 300px\" \/><\/p>\n<p>&ldquo;The heart needs a lot of energy to contract and to move blood around your body, so it gets that energy by using a lot of sugars, mostly fats and amino acids,&rdquo; Lee explained. &ldquo;We wanted to see if <abbr>PKM2<\/abbr> made processing glucose more efficient and if it helps maintain energy when resources are limited, like oxygen.&rdquo;<\/p>\n<p>Unexpectedly, Lee and the team found that, in the absence of <abbr>PKM2<\/abbr>, glucose was redirected from energy production to managing oxidative stress, which is often triggered by dysfunctional mitochondria.<\/p>\n<p>&ldquo;This has been a paper that we&#8217;ve been waiting for years to be published, and it&#8217;s been a journey to get all the right pieces together,&rdquo; Lee said. &ldquo;If anyone\u2019s interested in reading it, they\u2019ll find years worth of data. I\u2019m happy to push it out and move on to the next part!&rdquo;<\/p>\n<p>Lee\u2019s journey began as an undergraduate biochemistry student at <abbr>¶«¾«Ó°Òµ<\/abbr> M\u0101noa and continued through her involvement with <abbr>JABSOM<\/abbr> and the <a href=\"https:\/\/inbre.jabsom.hawaii.edu\/\">IDeA Networks of Biomedical Research Excellence <\/a> program.
